Betaine and carnitine standard/Sample solutions were derivatised with a mixture of 2-Bromo-2'-acetonaphthone and 18-crown-6 in N-methyl -2 pyrrolidone in presence of KH<sub>2</sub>PO<sub>4</sub> (0.1 M) at 80<sup>◦</sup>C (4 h). After filtering, the solution was eluted with 22 mM Choline chloride in 10% H<sub>2</sub>O/90% ACN at a flow rate of 1.9 ml/min (Injection 5 µL) at 40<sup>◦</sup>C. Betaine content in microalgae biomass was determined using calibration curve of standard betaine.<br>
甜菜碱(Betaine)与肉碱(carnitine)的标准溶液及样品溶液,在0.1 mol/L磷酸二氢钾(KH₂PO₄)存在条件下,于80℃中采用2-溴-2'-乙酰萘(2-Bromo-2'-acetonaphthone)与18-冠-6(18-crown-6)的混合液在N-甲基-2-吡咯烷酮(N-methyl-2-pyrrolidone)中进行衍生化反应,反应时长为4小时。过滤完成后,将所得溶液以1.9 mL/min的流速进行洗脱,洗脱液为含22 mM氯化胆碱(Choline chloride)的10%水/90%乙腈(ACN)混合溶剂,柱温设置为40℃,进样量为5 μL。采用甜菜碱标准品绘制校准曲线,对微藻生物质中的甜菜碱含量进行定量测定。
