Chromosome-level genome assembly of an endangered plant Prunus mongolica using PacBio and Hi-C technologies

<em>Prunus mongolica</em> is an ecologically and economically important xerophytic tree native to Northwest China. Here, we report a high-quality, chromosome-level <em>P. mongolica</em> genome assembly integrating PacBio high-fidelity sequencing and Hi-C technology. The assembled genome was 233.17 Mb in size, with 98.89% assigned to eight pseudochromosomes. The genome had contig and scaffold N50s of 24.33 Mb and 26.54 Mb, respectively, a BUSCO completeness score of 98.76%, and CEGMA indicated that 98.47% of the assembled genome was reliably annotated. The genome contained a total of 88.54 Mb (37.97%) of repetitive sequences and 23,798 protein-coding genes.

蒙古扁桃(Prunus mongolica)是一种兼具重要生态与经济价值的旱生乔木,原产于中国西北地区。本研究整合PacBio高保真测序(PacBio high-fidelity sequencing)与Hi-C技术(Hi-C technology),报道了一个高质量、染色体级别的蒙古扁桃基因组组装结果。组装完成的基因组总长度为233.17 Mb,其中98.89%的序列被锚定至8条假染色体上。该基因组的重叠群(contig)N50长度为24.33 Mb,支架(scaffold)N50长度为26.54 Mb;BUSCO完整度评分达98.76%,CEGMA分析显示98.47%的组装序列可被可靠注释。基因组中共包含88.54 Mb(占比37.97%)的重复序列,以及23798个蛋白质编码基因。
