Proteinuria in G1/G1 mice (raw data) treated with lisinopril and untreated

Here I studied the effect of lisinopril in the G1/G1 mouse model. Most G1/G1 mice survived the four-week experimental period with ongoing proteinuria and moderate to severe (morphologically defined) FSGS, but a few mice with more severe disease died between weeks two and four. This table represents pooled data from two separate experiments; one terminated at day 38, and another continued to day 60 to assess survival. 24 G1/G1 mice were injected with the pCpG free interferon ɤ plasmid. To exclude any injected mice that did not develop kidney disease, we confirmed the presence of proteinuria by SDS PAGE chromatography of urine at day seven post-injection and only proteinuric mice were randomized to either treatment or control groups (12 mice each). Lisinopril was administered in the drinking water at 75 mg/L, and the control group received water without drug. Urine was collected twice a week. The first group of mice was sacrificed for analysis on day 38 post-injection. Proteinuria (raw data) is expressed as ugAlbumin/mg Creatinine.

本研究针对G1/G1小鼠模型中赖诺普利(lisinopril)的干预效应展开探究。多数G1/G1小鼠可顺利度过为期4周的实验周期,期间持续表现出蛋白尿,并呈现中度至重度(经形态学定义的)局灶节段性肾小球硬化(focal segmental glomerulosclerosis, FSGS);但少数病情更严重的小鼠在实验第2至4周间死亡。本表格汇总自两项独立实验的数据:其中一项实验于注射后第38天终止,另一项实验持续至第60天以评估小鼠存活率。我们向24只G1/G1小鼠注射了无CpG干扰素γ质粒(pCpG free interferon γ plasmid)。为排除未发生肾脏病变的注射小鼠,我们于注射后第7天通过尿液的十二烷基硫酸钠-聚丙烯酰胺凝胶电泳(SDS-PAGE)色谱分析确认了蛋白尿的存在,仅将出现蛋白尿的小鼠随机分配至治疗组与对照组(每组各12只)。治疗组小鼠的饮用水中添加了75 mg/L的赖诺普利,对照组小鼠则饮用不含药物的普通饮水。每周采集两次尿液样本。第一组小鼠于注射后第38天被处死以进行相关检测分析。蛋白尿原始数据以微克白蛋白/毫克肌酐(ugAlbumin/mg Creatinine)为单位进行表述。
