Bacterial microbiota in nest and eggshell samples - Observational study

This study aim at testing the effect of aromatic plants on bacterial microbiota of nest samples in an experimental approach, and on bacterial microbiota of nest material and eggshell and on nestling traits and behavior in an observational approach in Corsican blue tits (<i>Cyanistes caeruleus ogliastrae</i>).The metadata and codes to analysis the data can be found in the following GitHub public repository: To add.<b>Observational approach</b>In spring 2023, we observe the quantity of aromatic plants that Corsican blue tits used as nest material in our study system along the reproductive season. We collected nest material during incubation and post-hatching, and eggshell swab during incubation. We extracted the DNA from the samples and amplified the 16S rARN sequences by PCR for the study of bacterial community. 本研究旨在通过实验方法探究芳香植物对科西嘉蓝山雀（*Cyanistes caeruleus ogliastrae*）巢样本细菌菌群的影响，并通过观测方法分析其对巢材、卵壳细菌菌群以及雏鸟性状与行为的影响。本研究的元数据与数据分析代码可在以下GitHub公共仓库获取：待补充。

**观测方法**
2023年春季，我们在研究样地中沿繁殖季监测了科西嘉蓝山雀用作巢材的芳香植物用量。分别于孵化期和孵化后期采集巢材样本，并于孵化期采集卵壳拭子样本。我们从样本中提取DNA，通过聚合酶链式反应（PCR）扩增16S rRNA序列以分析细菌群落，采用Illumina Mi-Seq平台对样本进行测序。

本数据集涵盖以下样本的细菌正向与反向测序读段：
1. 卵壳样本：46份卵壳拭子样本、3份野外阴性对照、2份DNA提取阴性对照、2份PCR阴性对照、2份PCR阳性对照
2. 巢材样本：
- 孵化期采集的巢材样本46份
- 孵化后期采集的巢材样本46份
- 3份野外阴性对照、1份DNA提取阴性对照、1份PCR阴性对照、1份PCR阳性对照
