Macrophages residing inside mouse pancreatic islets have an activated phenotype at steady-state. To explore the molecular pathways involved in their polarization, we compared the transcriptomic signatures of islet-resident macrophages with autologous epididymal adipose tissue (EpiWAT)-resident macrophages. Our data confirmed the pro-inflammatory profile of islet marophages and revealed also strong anti-oxidant capacities compared to EpiWATcells. Overall design: Single cells were obtained from pancreatic islets and EpiWAT of autologous 12-week-old chow diet-fed C57BL/6J males. Resident marcrophages were sort-purified as live DAPI-CD45+F4/80+CD64+ cells with a FACS ARIA III cell sorter (BD Biosciences) using FACS Diva software (BD Biosciences) and lysed for mRNA extraction and librairie preparation.
