Forward primers begin with sequence containing attB1 sites (underlined) and additional nucleotides (gg) to place following sequence in-frame with polyhistine coding sequence in ultimate expression plasmid followed by gene-specific target sequences (…xxx-3′).Reverse primers begin with sequence containing attB2 sites (underlined) followed by stop codon (tta), then gene-specific target sequence.正向引物的序列以带有下划线标注的attB1位点（attB1 site）作为起始序列，额外添加了两个核苷酸（gg），以使后续序列与最终表达质粒中的多聚组氨酸编码序列（polyhistidine coding sequence）保持读码框一致，随后连接基因特异性靶序列（…xxx-3′）。反向引物的序列以带有下划线标注的attB2位点（attB2 site）作为起始序列，随后接上序列为tta的终止密码子（stop codon），再连接基因特异性靶序列。
