Polymeric mesoporous carrier from cellulose acetate butyrate was loaded with Felodipine and molecular interactions between the drug and internal mesopore surface explored. The findings reveal that a ‘surface anchoring’ effect could be responsible for the lack of crystal-like ordered structure of the drug within mesopores. The more soluble amorphous form was preserved inside the carrier’s mesopores, despite its known poor glass forming ability, which translated into enhanced dissolution and relative bioavailability.

以醋酸丁酸纤维素制备的聚合物介孔载体负载了非洛地平(Felodipine),并对该药物与载体介孔内表面之间的分子相互作用展开了探究。研究结果表明,“表面锚定(surface anchoring)”效应是导致药物在介孔孔道内未形成类晶体有序结构的原因。尽管已知该药物本身成玻璃性能较差,但其溶解度更优的无定形形态仍能在载体介孔孔道内稳定保留,这一特性最终实现了药物溶出度与相对生物利用度的提升。
