Screening of differentially expressed circulating microRNAs during osteoporosis. Homo sapiens

To explore the variation of serum microRNA expression during osteoporosis, we have employed microRNA microarray expression profiling as a discovery platform to identify microRNAs with the potential to diagnose osteoporosis from healthy and osteopenia individuals for clinical use. Whole blood from healthy, osteopenic and osteoporotic donors was collected, and the sera were separated. Twenty two microRNAs (miR-15a-5p, miR-29b-5p, miR-30c-2-3p, miR-145-5p, miR-199a-5p, miR-301a-3p, miR-424-5p, miR-497-5p, miR-526b-5p, miR-550a-5p, miR-575, miR-654-5p, miR-663a, miR-708-5p, miR-877-3p, miR-1246, miR-1260b, miR-1299, miR-1323, miR-4447, miR-4769-3p and miR-5685) were finally used for further detection of osteoporosis diagnosis. Overall design: Gradually differed microRNAs were measured in healthy, osteopenic and osteoporotic participants. Three independent cohorts were performed to verify the diagnostic performance.
为探究骨质疏松症患者血清微小核糖核酸(microRNA)表达谱的变化,本研究采用微小核糖核酸微阵列表达谱分析作为发现平台,旨在筛选出可用于临床、区分健康人群、骨量减少个体与骨质疏松患者的诊断性微小核糖核酸。收集健康、骨量减少及骨质疏松供者的全血,分离得到血清。最终选取22种微小核糖核酸(miR-15a-5p、miR-29b-5p、miR-30c-2-3p、miR-145-5p、miR-199a-5p、miR-301a-3p、miR-424-5p、miR-497-5p、miR-526b-5p、miR-550a-5p、miR-575、miR-654-5p、miR-663a、miR-708-5p、miR-877-3p、miR-1246、miR-1260b、miR-1299、miR-1323、miR-4447、miR-4769-3p及miR-5685)用于后续骨质疏松诊断相关检测。整体实验设计:对健康、骨量减少及骨质疏松受试者体内的差异表达微小核糖核酸进行检测,设置3个独立队列以验证其诊断性能。
