烏心石舅活性成分Syringin及Caryophyllene之研究

本研究專注在台灣特有種烏心石舅(Magnolia kachirachirai)、中國大陸樂東擬單性木蘭( Magnolia lotungensis)及雲南烏心石舅(Magnolia yunnanensis )這三物種、種間和種內的主要活性成分分析,烏心石舅最主要之成分為Syringin、β-Caryophyllene及Caryophyllene oxide,在抗發炎活性分析方面,烏心石舅枝部位所分離出的15個分離部皆有不錯的細胞存活率及修復發炎反應,氣相層析質譜儀成分分析方面,烏心石舅具有較高的trans-β - Ocimene、(-)-β-Elemene、β-Caryophyllene、α-Bisabolene成分,這些成分皆用於香料工業上,Syringe含量方面,以烏心石舅Taimali 4單株、枝部位分離部Fraction 3含量較高,烏心石舅物種與中國大陸兩物種相比較Syringin含量高於樂東擬單性木蘭及雲南烏心石舅,薄層層析法成分分析方面,太麻里地區烏心石舅葉、枝及樹皮部位之Syringin含量,以編號太麻里11之含量較高,創傷後活性成分之定性及定量分析方面,RT (Residence time)65.11、65.14及65.17之成分為Caryophyllene,60分鐘後含量逐漸增加,主要是細胞開始進行修復功能,SPME分析方面,得到3個成分,RT 8.18(2-Pyrrolidinone)、RT 49.03(Cyclohexanone)及RT 96.00(Hydrazinecarboxamide)。

This study focuses on the analysis of major active components within and between three species: *Magnolia kachirachirai*, an endemic species to Taiwan, *Magnolia lotungensis* from mainland China, and *Magnolia yunnanensis*. The primary active components of *M. kachirachirai* are syringin, β-caryophyllene, and caryophyllene oxide. In anti-inflammatory activity assays, all 15 fractions isolated from the twig tissues of *M. kachirachirai* exhibited favorable cell viability and alleviated inflammatory responses. Through gas chromatography-mass spectrometry (GC-MS) analysis, *M. kachirachirai* was found to have relatively high contents of trans-β-ocimene, (-)-β-elemene, β-caryophyllene, and α-bisabolene, all of which are widely used in the flavor and fragrance industry. Regarding syringin content, the Taimali 4 individual of *M. kachirachirai* and Fraction 3 isolated from its twig parts showed relatively high syringin levels. Compared with the two mainland Chinese species, the syringin content of *M. kachirachirai* is higher than that of *M. lotungensis* and *M. yunnanensis*. Through thin-layer chromatography (TLC) analysis, among the leaf, twig, and bark tissues of *M. kachirachirai* collected from the Taimali region, the sample numbered Taimali 11 exhibited the highest syringin content. In qualitative and quantitative analysis of active components post-wounding, the components with retention times (RT) of 65.11, 65.14, and 65.17 were identified as caryophyllene, whose contents gradually increased 60 minutes after wounding, as cells began to initiate repair functions. Via solid-phase microextraction (SPME) analysis, three components were identified: RT 8.18 (2-pyrrolidinone), RT 49.03 (cyclohexanone), and RT 96.00 (hydrazinecarboxamide).
