We prospectively isolate and characterize first and second heart field- and nodal-like cardiomyocytes using a double reporter line from human embryonic stem cells Overall design: We performed RNA sequencing of hESC derived CMs at various timepoints, and scRNA seq on Day 20 hESC derived CMs and human fetal heart at 10 weeks of gestation.

本研究采用人类胚胎干细胞(human embryonic stem cells)来源的双报告基因系,前瞻性地分离并鉴定第一心区(first heart field)、第二心区(second heart field)及结样心肌细胞(nodal-like cardiomyocytes)。整体实验设计:我们对不同时间点的人类胚胎干细胞诱导心肌细胞(hESC-derived cardiomyocytes, CMs)进行RNA测序,并对第20天的人类胚胎干细胞诱导心肌细胞以及妊娠10周的人类胎儿心脏开展单细胞RNA测序(scRNA seq)。
