Ewing’s sarcoma (EWS) is a cancer of the bones or soft tissues in children and adolescents. EWS-FLI1 is a transcriptional factor and the key driver of EWS. To characterize the changes of downstream transcriptional profiles of EWS-FLI1 in A673 cells upon knockdown of EWS-FLI1 and deubiquitinase USP9X, RNA-seq was performed in A673 cells transfected with EWS-FLI1 esiRNA, USP9X esiRNA or GFP esiRNA as control. The data indicates that USP9X regulates transcriptional profiles of EWS through mediating EWS-FLI1. Overall design: Transcriptional profile of A673 cells upon USP9X knockdown and EWS-FLI1 knockdown.
尤因肉瘤(Ewing’s sarcoma, EWS)是一类好发于儿童与青少年的骨骼或软组织恶性肿瘤。EWS-FLI1属于转录因子,亦是尤因肉瘤发生发展的关键驱动因子。为解析A673细胞中,分别敲低EWS-FLI1或去泛素化酶USP9X(deubiquitinase USP9X)时,EWS-FLI1下游转录谱的变化,研究人员对转染了EWS-FLI1 esiRNA、USP9X esiRNA以及作为对照的GFP esiRNA的A673细胞开展了RNA测序(RNA-seq)。实验数据表明,USP9X可通过介导EWS-FLI1的方式调控尤因肉瘤的转录谱。整体实验设计:分析A673细胞分别在USP9X敲低与EWS-FLI1敲低条件下的转录谱特征。
