Investigation of TOF patient heart specimens

The protein BBLN with largely unknown function was identified in human right ventricular heart specimens from patients undergoing repair surgery for Tetralogy of Fallot (TOF). Proteins from heart specimens were solubilized and subjected to immuno-affinity enrichment with anti-CAMK2D-specific antibodies. Co-enriched proteins were separated by SDS-PAGE, and the protein band in the range of 8-14 kDa was cut and subjected to identification by nano-LC-ESI-MS/MS.功能尚未明确的蛋白BBLN，在接受法洛四联症（Tetralogy of Fallot，TOF）修复手术的患者的人类右心室心脏标本中被鉴定得到。研究人员对心脏标本中的蛋白进行增溶处理，随后使用抗CAMK2D特异性抗体开展免疫亲和富集。共富集得到的蛋白经十二烷基硫酸钠-聚丙烯酰胺凝胶电泳（SDS-PAGE）分离，切取分子量在8~14 kDa范围内的蛋白条带，通过纳米液相色谱-电喷雾串联质谱（nano-LC-ESI-MS/MS）进行鉴定。
