A large-scale analysis of histone acetylation as well as RNA polymerase II and GATA 1 interactions on chromosome 6q in human erythroid progenitor cells. Human erythroid progenitors from day 10 erythroid cultures were used for ChIP-chip analysis. For this purpose, the NimbleGen array system was used, following manufacturer's protocols. Samples for ChIP were taken from the same individual and processed for use with each of the following antibodies: GATA1, PolII, AcH3 and AcH4.

本研究针对人类红系祖细胞中6号染色体长臂(chromosome 6q)区域的组蛋白乙酰化、RNA聚合酶II(RNA polymerase II)与GATA结合蛋白1(GATA1)的相互作用开展大规模分析。实验选取第10天红系培养物中的人类红系祖细胞,用于染色质免疫沉淀芯片(ChIP-chip)分析。本次实验遵循生产商的操作规范,采用NimbleGen阵列系统完成检测。染色质免疫沉淀所用样本均取自同一供体,并分别使用以下抗体进行处理:GATA1、PolII、乙酰化组蛋白H3(AcH3)以及乙酰化组蛋白H4(AcH4)。
