To unedrstand the global gene expression profile depending upon origin of inflammation, we have employed microarray analysis in vitro. We have treated THP-1 macrophage like cells with TNFα, IL-6 to mimic inflammatory milieu similar to systemic inflammation while bacterial LPS treatment represented for infectious inflammation. THP-1 macrophage like cells were treated with TNFα (10 ng/ml), IL-6 (50 ng/ml) and LPS (100 ng/ml) for 2 hours. Cells were treated with PBS served as control

为探究基于炎症起源的全局基因表达谱,我们采用体外(in vitro)微阵列分析(microarray analysis)技术。我们使用肿瘤坏死因子α(TNFα)、白细胞介素6(IL-6)处理THP-1巨噬细胞样细胞(THP-1 macrophage-like cells),以模拟与全身性炎症相似的炎症微环境;而细菌脂多糖(LPS)处理则代表感染性炎症模型。实验中分别以10 ng/ml的TNFα、50 ng/ml的IL-6及100 ng/ml的LPS处理细胞,处理时长为2小时。以磷酸盐缓冲液(PBS)处理的细胞作为空白对照。
