We performed scRNA-seq to assess the effect of CpG-STAT3ASO+anti-PD1 therapy on immune cells from GL261 high grade glioma model. Overall design: The mice were implanted with GL261 cells. Subsequently, the mice received four intracranial treatments with either PBS, CpG-STAT3ASO, or CpG-STAT3ASO combined with anti-PD1, and were sacrificed on day 21. Tumor-bearing hemispheres were then isolated and dissociated. CD45+ cells were sorted and subjected to single-cell RNA sequencing (10X Genomics 3').

本研究通过单细胞RNA测序(single-cell RNA sequencing, scRNA-seq),评估CpG-STAT3ASO联合抗PD-1(anti-PD1)疗法对GL261高等级胶质瘤模型来源免疫细胞的影响。总体实验设计:将GL261细胞接种至小鼠体内;随后,小鼠分别接受PBS、CpG-STAT3ASO、CpG-STAT3ASO联合抗PD-1(anti-PD1)四种颅内给药处理,共给药四次,并于接种后第21天安乐死。分离荷瘤大脑半球并进行组织解离,分选CD45阳性细胞后,采用10X Genomics 3'端单细胞RNA测序平台完成测序。
