argeted sequencing of barcodes from genomic DNA harvested from barcoded human fibroblast-like cells (hiF-T cells) and human induced pluripotent stem cells (hiPS cells) derived from hiF-T cells. hiF-T cells were transduced with barcode libraries consisting of semi-random 100-mers in the 3' UTR of GFP and permitted to undergo several cell divisions before being split and seeded for parallel reprogrammings. Both barcoded hiF-T cells and hiPS cells were harvested for isolation of genomic DNA before we performed targeted amplification of the barcode vector for sequencing.
