In this study, we used next-generation sequencing technologies and tandem mass tags to characterize mRNA-seq, miRNA-seq and proteomic of Pelteobagrus fulvidraco, P. vachelli and hybrid yellow catfish Huangyou-1 (P. fulvidraco female×P. vachelli male) livers and in doing so, offer deeper insight into the transcriptional and protein changes in heterosis uncovers key roles for miRNAs. 本研究采用下一代测序技术与串联质量标签(tandem mass tags, TMT)技术,对黄颡鱼(Pelteobagrus fulvidraco)、瓦氏黄颡鱼(Pelteobagrus vachelli)及其杂交品种黄优1号(P. fulvidraco ♀ × P. vachelli ♂)的肝脏组织开展mRNA测序(mRNA-seq)、微小RNA测序(miRNA-seq)以及蛋白质组学分析;借此深入解析杂种优势相关的转录与蛋白水平变化,并揭示微小RNA在此过程中的关键调控作用。实验设计概述:参照厂商推荐的实验流程,使用Illumina HiSeq 2500测序平台(美国LC Sciences公司)完成RNA测序分析。
