Bulk RNA sequencing of R. parkeri infected and uninfected hemocytes from Amblyomma maculatum revealed 39,249 mRNA transcripts. Differential gene expression analysis revealed 17 upregulated and 35 downregulated genes with immune functions in R. parkeri infected hemocytes. Using a reverse genetic approach (RNAi), we characterized the functions of two genes, nimrod B2, and eater as unique to phagocytic hemocytes and critical for phagocytosis.

对美洲钝眼蜱(Amblyomma maculatum)的帕克氏立克次体(R. parkeri)感染组与未感染组的血细胞开展批量RNA测序(bulk RNA sequencing),共鉴定得到39249条mRNA转录本。经差异基因表达分析,在R. parkeri感染的血细胞中发现17个免疫相关上调基因与35个免疫相关下调基因。本研究借助反向遗传学方法(RNA干扰,RNAi),对nimrod B2与eater两个基因的功能进行了表征,证实二者为吞噬性血细胞所特有,且对吞噬作用至关重要。
