To study the community composition of picoeukaryotes during successive Noctiluca and Mesodinium blooms, 500 mL surface water samples were collected from Port Shelter, Hong Kong, pre-filtered through a 200 μm mesh, and further filtered through a 3.0 µm (47 mm) polycarbonate membrane (PALL Corporation). Then water samples were filtered onto a 0.22 µm (47 mm) polycarbonate membrane for DNA extraction (GE Water and Process Technologies), using a vacuum pump at 0.034 bar Hg or below. The filters were flash frozen and stored at -80ºC until DNA extraction. Samples were then studied using 454-pyrosequencing

为探究夜光藻(Noctiluca)与中缢虫(Mesodinium)相继水华期间的超微型真核生物(picoeukaryotes)群落组成,研究人员从香港吐露港(Port Shelter)采集500 mL表层海水样本。样本先经200 μm孔径网筛预过滤,再依次通过3.0 μm(47 mm)聚碳酸酯滤膜(颇尔公司,PALL Corporation)过滤。随后将水样经0.22 μm(47 mm)聚碳酸酯滤膜(GE水处理及工艺技术公司,GE Water and Process Technologies)截留,采用真空泵维持0.034 bar汞柱及以下的真空度进行抽滤以用于DNA提取。滤膜经快速冷冻后保存于-80℃环境,直至开展DNA提取操作。最终采用454焦磷酸测序技术(454-pyrosequencing)对样本进行分析。
