Sequencing clinical isolates from dental plaque. Streptococcus mutans TCI-149

Plaque samples were collected from twin children with and without dental caries. S. mutans and S. sobrinus were isolated by plating plaque scrapings on selective media. Unique isolates were chosen for genome sequencing by restriction digest genotyping; libraries were constructed using the EpiBio Nextera DNA sample prep kit, and sequencing was performed using an Illumina Genome Analyzer IIx.本研究从伴龋病与无龋病的双胞胎儿童中采集牙菌斑样本。通过将牙菌斑刮取物涂布于选择性培养基进行培养，分离得到变形链球菌（Streptococcus mutans，简称S. mutans）与远缘链球菌（Streptococcus sobrinus，简称S. sobrinus）。经限制性酶切基因分型筛选出独特分离株用于全基因组测序；使用EpiBio Nextera DNA样本制备试剂盒构建测序文库，并采用Illumina Genome Analyzer IIx测序平台完成测序。
