RNA sequencing was performed to compare glomerular transcriptomes of control and podocyte-specific DACH1 KO mice both under the basal condition and early after STZ-induced diabetes. Glomerular RNA was extracted from ten week old age-matched control and podocyte-specific DACH1 KO mice under the basal condition and RNA sequencing performed. Control and podocyte-specific DACH1 KO mice were also made diabetic by STZ injection and ten days later, glomerular RNA was isolated and RNA sequencing performed. Three different mice from each experimetal group were analyzed.
本研究采用RNA测序(RNA Sequencing)技术,对比分析基础状态与链脲佐菌素(Streptozotocin,STZ)诱导糖尿病早期两个条件下,对照组及足细胞(podocyte)特异性DACH1敲除(Knock Out,KO)小鼠的肾小球转录组。首先从10周龄、年龄匹配的基础状态对照组小鼠与足细胞特异性DACH1 KO小鼠的肾小球组织中提取RNA并开展RNA测序;此外,通过STZ注射诱导对照组与足细胞特异性DACH1 KO小鼠发生糖尿病,造模10天后分离其肾小球RNA并进行RNA测序。每个实验组均选取3只不同的小鼠进行分析。
