In this study, we generated a randomly barcoded transposon mutant library in Sphingobium sp. SYK-6 and applied it for characterization of lignin-related aromatic catabolism. TnSeq mapped unique barcoded transposon insertions to specific genomic loci, and then BarSeq enumerated the number of barcode insertions before and after various growth selection experiments.
