RB-TnSeq in Sphingobium sp. SYK-6

In this study, we generated a randomly barcoded transposon mutant library in Sphingobium sp. SYK-6 and applied it for characterization of lignin-related aromatic catabolism. TnSeq mapped unique barcoded transposon insertions to specific genomic loci, and then BarSeq enumerated the number of barcode insertions before and after various growth selection experiments.本研究在鞘氨醇单胞菌属（Sphingobium sp.）SYK-6菌株中构建了随机条形码标记的转座子突变体库，并将其用于木质素相关芳香族化合物分解代谢的特性解析。转座子测序（TnSeq）可将唯一条形码标记的转座子插入位点定位至特定基因组位点，随后通过条形码测序（BarSeq）统计了各类生长筛选实验前后的条形码插入拷贝数。
