MiR-34a and miR-34b/c encoding genes represent direct targets of the p53 tumor suppressor. MiR-34a/b/c mediate tumor suppression by p53 via down-regulation of the expression of specific target mRNAs. Here, we characterized the miR-34 effectors using a CRISPR/Cas9-approach in HCT116 cells with or without 5-FU treatment. HCT116 wildtype cells and HCT116 miR-34a/b/c-KO cells were treated with DMSO or 5-FU for 48 hours. For each group, RNAs from three biological replicates were obtained.

编码miR-34a与miR-34b/c的基因,是p53肿瘤抑制因子的直接作用靶标。miR-34a/b/c可通过下调特定靶标mRNA的表达,介导p53所发挥的肿瘤抑制作用。本研究采用CRISPR/Cas9技术,在经或未经5-氟尿嘧啶(5-FU)处理的HCT116细胞中,对miR-34的效应因子进行了功能表征。将HCT116野生型细胞与HCT116 miR-34a/b/c基因敲除细胞分别用二甲基亚砜(DMSO)或5-氟尿嘧啶处理48小时。每组均获取三份生物学重复的RNA样品。
