We conducted Ribo-Zero RNA-Seq studies on rat hippocampus to elucidate changes in RNA expression in the hippocampus under physiological conditions such as acute stress and circadian variation. Rats were subjected to 15-min forced swim (FS) session and hippocampus tissue was collected at 30, 60, 120, 180 or 360 minutes after the onset of swimming. Tissues were also collected under early morning baseline (BL AM) or late afternoon baseline (BL PM) conditions. Based on this analysis, we have identified many genes that are differentially expressed as a result of acute stress exposure and circadian variation. Overall design: 37 Ribo-Zero RNASeq samples across 7 conditions: 5 biological replicates per condition for BLAM, BLPM, FS120, FS180, FS360; and 6 biological replicates for FS30 and FS60

本研究针对大鼠海马体开展Ribo-Zero RNA测序(Ribo-Zero RNA-Seq)实验,旨在阐明急性应激、昼夜节律变化等生理条件下海马体的RNA表达变化规律。将大鼠实施15分钟强迫游泳(FS)处理,并于游泳启动后的30、60、120、180或360分钟采集其海马体组织;同时还在清晨基线(BL AM)与傍晚基线(BL PM)条件下采集海马体组织。基于本次测序分析,本研究已鉴定出大量因急性应激暴露与昼夜节律变化而发生差异表达的基因。实验整体设计:共计37份Ribo-Zero RNA测序样本,涵盖7种实验条件:BL AM、BL PM、FS120、FS180、FS360每组各设置5个生物学重复,FS30与FS60每组各设置6个生物学重复。
