We used a rat model of whole body (except one hind limb that was shielded) x-ray irradiation to profile the microRNA (miRNA) in kidneys at 35 days after radiation. Small RNA from normal and irradiated (with or without lisinopril) Wistar rat kidneys were analyzed by next-generation sequencing and the changes by radiation and lisinopril were identified by deRNA-seq. MiR-34a-5p was increased after irradiation. Overall design: Profile of miRNA in Wistar rat kidneys with and without radiation at 35 days after radiation were generated by next generation sequencing. Rats were randomized into 3 groups (1) nonirradiated controls (0 Gy). (2) Irradiation only group (13 Gy). (3) Irradiation + lisinopril group (13 Gy + lisinopril). Each group contains four or five rats.
本研究采用全身(单侧后肢受屏蔽)X射线照射的大鼠模型,于照射后35天对肾脏中的微小RNA(microRNA,miRNA)开展表达谱分析。提取正常及照射处理(给予或不给予赖诺普利)的Wistar大鼠肾脏中的小RNA,通过下一代测序(next-generation sequencing)进行分析,并借助deRNA-seq鉴定辐射与赖诺普利所引发的表达变化。MiR-34a-5p的表达水平在照射后上调。
实验整体设计:通过下一代测序获取照射后35天的Wistar大鼠肾脏中的miRNA表达谱。将大鼠随机分为3组:(1) 非照射对照组(0 Gy);(2) 单纯照射组(13 Gy);(3) 照射+赖诺普利组(13 Gy + 赖诺普利)。每组包含4~5只大鼠。
