To investigate the functional difference between rs1260326:C>T in the regulation of lipogenesis in the liver, we established human induced pluripotent stem cell lines from GCKR CC and GCKR TT variants. Overall design: We then performed gene expression profiling analysis using data obtained from RNA-seq of human liver organoids (HLOs) generated from 5 different cell lines of which 2 are GCKR TT variants and 3 from GCKR CC variants Comparative gene expression profiling analysis of RNA-seq data for human liver organoids generated from GCKR TT and CC cell lines.

为探究rs1260326:C>T位点在肝脏脂肪生成调控中的功能差异，本研究从携带GCKR CC与GCKR TT基因型的细胞中建立了人诱导多能干细胞（human induced pluripotent stem cell, hiPSC）系。实验整体设计：我们采用5株不同细胞系来源的人肝脏类器官（human liver organoids, HLOs）的RNA测序（RNA-seq）数据开展基因表达谱分析，其中2株为GCKR TT基因型细胞系，剩余3株为GCKR CC基因型细胞系；并针对上述GCKR TT与CC基因型细胞系来源的人肝脏类器官的RNA测序数据完成了对比基因表达谱分析。
