The gole of this project was to explore the effect of NH3 and/or H2S effect on the mouse sperm small RNA expression and the trangerational effects. small RNA sequencing of mouse sperm samples from different groups (NH3 and or H2S treatment) in different generations was done to search for transgenerational effects on RNA expression. small RNA seqencing of mouse sperm samples from different groups in different generations to search for transgenerational effects of NH3 and/or H2S treatment. First, three weeks of age ICR male mice were treated with NH3 and H2S for 5 weeks (F0 mice). Then the mice (F0) mated with normal ICR female mice. After delivery, F1 male mice were raised normally (without any treatments). At the 8 weeks of age, F1 male mice mated with normal female mice. The F2 male mice were raised normally without any treatment. The semen was collected for F0, F1, F2 mice for the RNA-seq to search for the transgenerational effects of NH3, H2S.

本项目旨在探究氨气(NH3)和/或硫化氢(H2S)对小鼠精子小RNA(small RNA)表达的影响及其跨代遗传效应。研究通过对不同代次、经NH3或H2S处理的小鼠精子样本开展小RNA测序,以搜寻RNA表达相关的跨代遗传效应。具体实验流程如下:首先选取3周龄的ICR品系雄性小鼠,对其进行为期5周的NH3与H2S处理(记为F0代小鼠);随后将F0代小鼠与正常ICR雌性小鼠交配。分娩后,F1代雄性小鼠正常饲养(不施加任何处理),待其8周龄时与正常雌性小鼠交配;F2代雄性小鼠同样正常饲养,不施加任何处理。最终收集F0、F1、F2代小鼠的精液进行RNA测序(RNA-seq),以探究NH3和/或H2S处理的跨代遗传效应。
