Bt Cry toxin resistance mapping in Helicoverpa zea F2 crosses

The goal of this study was to use genetic crosses between Helicoverpa zea Cry toxin resistant and susceptible parents to identify genomic regions associated with Cry resistance. Growth of F2 offspring was measured on diets containing Cry1Ab and Cry1A.105 + Cry2Ab2 expressing leaf tissue and control diets containing non-Bt expressing leaf tissue. We used ddRAD sequencing of parents and offspring from these genetic crosses to generate markers across the genome for association with growth phenotype.
本研究旨在通过美洲棉铃虫(Helicoverpa zea)Cry毒素(Cry toxin)抗性品系与敏感品系间的遗传杂交,鉴定与Cry抗性相关的基因组区域。我们将F2代子代分别饲养于两类饲料:一类为添加表达Cry1Ab、Cry1A.105与Cry2Ab2的叶片组织的饲料,另一类为添加不表达Bt毒素的叶片组织的对照饲料,并测定其生长状况。随后,我们对上述遗传杂交的亲本及子代进行双酶切限制性位点相关DNA测序(double digest restriction-site associated DNA sequencing,ddRAD),以生成覆盖全基因组的分子标记,用于生长表型的关联分析。
