RNA of murine macrophages and neutrophils treated with LPS and VISTA

V-domain immunoglobulin suppressor of T-cell activation (VISTA) has emerged as a unique immunoregulatory receptor on cells of the myeloid lineage. Agonizing VISTA on myeloid cells has recently been demonstrated to have a profound effect on dampening inflammatory responses. VISTA has been proposed to function both as a ligand and as a receptor, yet the role of VISTA as a ligand has been largely ignored. In this experiment, macrophages and neutrophils were isolated from mice and treated with LPS and VISTA to examine the effect of VISTA in dampening acute inflammation.
T细胞活化免疫球蛋白V结构域抑制因子(V-domain immunoglobulin suppressor of T-cell activation,VISTA)现已被鉴定为髓系细胞表面一类独特的免疫调节受体。近期研究证实,激动髓系细胞表面的VISTA可对炎症反应产生显著的抑制作用。VISTA被认为既可作为配体也可作为受体发挥功能,但目前其作为配体的功能在很大程度上被忽视了。本实验中,研究人员从小鼠体内分离出巨噬细胞与中性粒细胞,分别用脂多糖(Lipopolysaccharide,LPS)与VISTA进行处理,以探究VISTA对急性炎症的抑制效果。
