Expression data of Gr-MDSCs under different treatment

Myeloid derived suppressor cells (MDSCs) are a heterogeneous population of immature myeloid cells with differentiation program partially blocked in pathological conditions such as cancer. Through inhibition of T cells especially cytotoxic T lymphocytes (CTLs), MDSCs play important tumor-promoting roles and maintain a state of immunological anergy and tolerance We used microarrays to profile gene expression of Gr-MDSC and identified differentially altered genes under different drug treatment. For 8-12 weeks old PB-Cre+ PtenL/L p53L/L Smad4L/L mTmGL/+ LSL-LucL/+ (CPPSML) male mice, once the prostate tumor reached 150mm3, the mice were surgically castrated and switched to enzalutamide-admixed diet for 4 weeks. The mice were then euthanized for isolating Gr-MDSCs from prostate tumors. Gr-MDSCs were then seeded into well-plate and treated with Dactolisib, Dasatinib or MDSC as control. RNA was isolated after 6h treatment and prepared for microarray.
髓系来源抑制细胞(myeloid derived suppressor cells, MDSCs)是一类异质性未成熟髓系细胞群,在癌症等病理条件下其分化程序会部分受阻。通过抑制T细胞,尤其是细胞毒性T淋巴细胞(cytotoxic T lymphocytes, CTLs),MDSCs可发挥重要的促肿瘤作用,并维持免疫无能与免疫耐受状态。我们利用微阵列(microarrays)对Gr-MDSC的基因表达谱进行分析,筛选出不同药物处理下的差异表达基因。实验所用对象为8-12周龄的PB-Cre+ PtenL/L p53L/L Smad4L/L mTmGL/+ LSL-LucL/+(CPPSML)雄性小鼠,当前列腺肿瘤体积达到150mm³时,对小鼠实施手术去势,并更换为掺有恩扎卢胺(enzalutamide)的饲料喂养4周。随后对小鼠实施安乐死,从前列腺肿瘤中分离Gr-MDSCs。将分离得到的Gr-MDSCs接种至培养板,分别用达科利西布(Dactolisib)、达沙替尼(Dasatinib)处理,以未处理的MDSC作为对照。处理6小时后提取RNA,制备样品用于微阵列实验。
