To detect the newly spacer sequence, the leader-proximal region of CRISPR array 1 was PCR amplified using Taq polymerase with forward primer Locus-F and reverse primer CRISPR1S4-R located at the 4th spacer of CRISPR array 1.The purified PCR products per sample was used for high-throughput sequencing.
为检测新型间隔序列(spacer sequence),我们采用Taq DNA聚合酶(Taq polymerase),以正向引物Locus-F以及结合于CRISPR阵列1第4个间隔区的反向引物CRISPR1S4-R,对CRISPR阵列1(CRISPR array 1)的前导序列近端区域进行PCR扩增。将每份样品的纯化PCR产物用于高通量测序(high-throughput sequencing)。
