A549 Lung Adenocarcinoma cells were engineered to co-express a PCNA chromobody tagged with mNeonGreen (green) and a truncated version of 53BP1 tagged with mApple (red). Cells were synchronised with 1mM thymidine and pulsed with 5µg/ml cisplatin for 2 hr prior to release to enrich cells in early S phase. Cells were then imaged using a Leica SP8 confocal microscopy, using HyD detectors a WLL and 63X 1.4 NA objective.
