Neoepitopes, tumor-specific antigens that arise from somatic mutations, are increasingly recognized as critical cancer immunotherapy targets. Thus, identifying immunogenic neoepitopes allows the assessment of endogenous anti-tumor responses and identifying clinically relevant immunotherapies. The aim of this project was to identify novel neoepitope candidates in the A20 murine B cell lymphoma cell line, through whole exome and RNA sequencing. By using ex vivo assays we identified immunogenic neoepitopes and showed that neoepitope-specific T cell responses can be induced in vivo by in situ vaccination in the A20 murine lymphoma model.
新表位(Neoepitopes)是源自体细胞突变的肿瘤特异性抗原,如今愈发被视作癌症免疫治疗的关键靶点。因此,鉴定具有免疫原性的新表位,可用于评估内源性抗肿瘤应答,并筛选具有临床价值的免疫治疗方案。本研究旨在通过全外显子组测序与RNA测序,在A20小鼠B细胞淋巴瘤细胞系中筛选新型新表位候选物。通过体外实验,本研究成功鉴定出免疫原性新表位,并证实:在A20小鼠淋巴瘤模型中,可通过原位疫苗接种在体内诱导新表位特异性T细胞应答。
