The aim of this research is to uncover novel post-transcriptional mechanisms regulating IL6 3'UTR. IL6 is one of the most essential cytokines and its expression is tightly controlled during acute and chronic inflammation. IL6 mRNA is known to be regulated by a variety of RNA binding proteins to adequately control immune response. However, our understanding of posttranscriptional regulation of this gene is still incomplete. To comprehensively address this issue, we performed a genome-wide screen to identify posttranscriptional regulators of IL6 3 prime UTR using CRISPR gRNA library. This study will provide us with a comprehensive list of potential regulators of IL6 3 prime UTR.
本研究旨在揭示调控白细胞介素6(Interleukin 6,IL6)3'非翻译区(3' Untranslated Region,3'UTR)的新型转录后(post-transcriptional)调控机制。IL6是最为关键的细胞因子之一,其表达在急性与慢性炎症进程中受到严格调控。已知IL6的mRNA可通过多种RNA结合蛋白(RNA binding protein)进行调控,以精准控制免疫应答。然而,目前学界对该基因的转录后调控机制的认知仍不全面。为全面解决这一问题,本研究利用CRISPR向导RNA(guide RNA, gRNA)文库开展全基因组筛选,以鉴定调控IL6 3'非翻译区的转录后调控因子。本研究将提供一份涵盖IL6 3'非翻译区潜在调控因子的完整列表。
