We conducted sequencing analysis of retrotransposon insertion sites for linkage map construction in sweetpotao. Cl8 was identified as a novel retrotransposon family which shows high insertion polymorphisms among sweetpotato cultivars. These insertion sites can be utilized as a molecular marker. Thus, we sequenced a number of Cl8 retrotransposon insertion sites using F1 mapping populations derived from two sweetpotato cultivars via Illumina high-throughput sequencing platform. Using those insertion sites, we constructed high-density linkage maps in sweetpotato.

本研究针对甘薯连锁图谱构建开展反转录转座子(retrotransposon)插入位点测序分析。Cl8被鉴定为一个新型反转录转座子家族,在甘薯栽培品种间呈现出较高的插入多态性。此类插入位点可作为分子标记加以利用。因此,本研究借助基于两个甘薯栽培品种构建的F1作图群体,通过Illumina高通量测序平台对多个Cl8反转录转座子插入位点进行测序。依托这些插入位点,本研究成功构建了甘薯高密度连锁图谱。
