Transcriptomeic reveal how green tea polyphenols against the Eimeria tenella infection in Wuliangshan black-bone chickens

Wuliangshan black-bone chickens were randomly divided into 5 groups with 15 chickens in each group. The chickens in 1-3 groups were treated with green tea polyphenols according to the body weight as follows: orally treated the total dose was 50mg water solution per weight kilogram (group 1), orally treated the total dose was 200mg water solution per weight kilogram (group 2), orally treated the total dose was 350mg water solution per weight kilogram (group 3). By contrast, group 4 was not treated with any tea polyphenols, whereas group 5 was the blank control group. The chickens were treated with green tea polyphenols from the age of 8 days to 28 days. The chickens in group1 to group 4 were orally infected with 50,000 sporulated E. tenella oocysts at 21 days old. At 28 days old, according to the ACI indexes, we choose the caecum of group 2 (green tea polyphenols treated and infected with Eimeria tenella), group 4 (infected with Eimeria tenella) and group 5 (not treated) for RNA-seq and validated differentially expressed genes.
本研究将无量山乌骨鸡随机分为5组,每组15只。第1至3组雏鸡按体重梯度给予绿茶多酚干预,具体方案如下:组1每千克体重经口灌服50mg绿茶多酚水溶液,组2每千克体重经口灌服200mg绿茶多酚水溶液,组3每千克体重经口灌服350mg绿茶多酚水溶液。组4未给予任何茶多酚处理,组5为空白对照组。所有受试鸡的绿茶多酚干预周期为8日龄至28日龄。组1至组4的雏鸡于21日龄时经口感染50000个柔嫩艾美耳球虫(Eimeria tenella, E. tenella)孢子化卵囊。于28日龄时,基于抗球虫指数(Anticoccidial Index, ACI),选取组2(经绿茶多酚干预并感染柔嫩艾美耳球虫)、组4(感染柔嫩艾美耳球虫)及组5(未处理组)的盲肠组织进行RNA测序(RNA-seq),并对差异表达基因(differentially expressed genes, DEGs)进行验证。
