Tissue-specific RNA profiling of the Brassica napus funiculus and funicular subcompartments using laser microdissection and RNA-sequencing Overall design: Funiculi were collected seven days after fertilization, histologically processed, and and embedded in paraffin. Entire funiculus or funiculus subcompartment (vasculature, epidermis, cortex) was collected using laser microdissection. RNA was extracted from laser microdissected tissues. This was used to prepare RNA-sequencing libraries with the Ovation RNA seq V2 cDNA amplificationand Illumina Truseq V2 library preparation kits.
基于激光显微切割(laser microdissection)与RNA测序(RNA-sequencing)的甘蓝型油菜(Brassica napus)珠柄(funiculus)及其亚分区组织特异性RNA谱分析 总体实验设计:于受精后7天收集珠柄,经组织学处理后进行石蜡包埋;通过激光显微切割获取完整珠柄或珠柄亚分区(维管组织、表皮、皮层);从激光显微切割得到的组织中提取RNA,并使用Ovation RNA seq V2 cDNA扩增试剂盒与Illumina Truseq V2文库制备试剂盒构建RNA测序文库。
