Rat skeletal muscle derived stem cells were grown in the presence of cholesterol or sodium palmitate in 1 mM glucose (low) or 4 mM glucose (high) RNA was extracted using the RNeasy Plus Micro kit and analyzed at UCLA by the Affymetrix rat Gene 2.0 ST chip. Muscle derived stem cells from male rats were isolated and grown. Cells in six well plates were subjected to 100 mg/dl cholesterol or sodium palmitate. RNA was isolated using the Qiagen RNeasy Plus Micro kit and subjected to analysis by DNA microarray using the Affymetrix rat Gene 2.0 ST chip.

将大鼠骨骼肌来源干细胞(rat skeletal muscle derived stem cells)置于添加了胆固醇(cholesterol)或棕榈酸钠(sodium palmitate),且分别含1 mM葡萄糖(低糖)或4 mM葡萄糖(高糖)的培养基中培养;随后使用RNeasy Plus Micro试剂盒(RNeasy Plus Micro kit)提取RNA,由加州大学洛杉矶分校(UCLA)采用Affymetrix大鼠Gene 2.0 ST芯片(Affymetrix rat Gene 2.0 ST chip)完成检测分析。分离雄性大鼠来源的骨骼肌干细胞并进行培养,将六孔板(six well plates)中的细胞以100 mg/dl浓度的胆固醇或棕榈酸钠处理,使用Qiagen RNeasy Plus Micro试剂盒提取RNA,再通过Affymetrix大鼠Gene 2.0 ST芯片开展DNA微阵列(DNA microarray)分析。
