Human corneal endothelial cells (HCEC) form a monolayer by adhering tightly through their intercellular adhesion molecules. Located at the posterior corneal surface, they maintain corneal translucency by dehydrating the corneal stroma, mainly through the Na+- and K+-dependent ATPase (Na+/K+-ATPase). Because HCEC proliferative activity is low in vivo,we tried to activate proliferation of HCEC by inhibiting cyclin-dependent kinase inhibitors.We have here demonstrated microarray data of transduced human corneal endothelial cell lines. Affymetrix human U133 plus 2.0 array was used to transcriptionally profile to compare cultured human corneal endothelial cells and transduced human corneal endothelial cells.

人角膜内皮细胞(human corneal endothelial cells,HCEC)可通过细胞间黏附分子紧密黏附形成单层细胞结构。该细胞定位于角膜后表面,主要依靠Na+和K+依赖性ATP酶(Na+/K+-ATPase)促使角膜基质脱水,进而维持角膜透明度。由于体内环境下人角膜内皮细胞的增殖活性极低,本研究尝试通过抑制细胞周期蛋白依赖性激酶抑制剂来激活其增殖能力。本次研究现已呈现转导后人角膜内皮细胞系的微阵列数据集,采用Affymetrix人类U133 plus 2.0基因芯片开展转录谱分析,对培养的人角膜内皮细胞与转导后人角膜内皮细胞进行对比。
