Overall design: Gene expression analysis of DMSO, single and combination treatment (ATRA and GSK2879552) on 6 AML cell lines at two time-points with two replicates (paired end RNA-seq on 96 samples in total)

全反式维甲酸(all-trans retinoic acid, ATRA)与GSK2879552联合处理,在基因表达、细胞增殖、分化标志物以及最为关键的细胞毒性层面均展现出协同效应。整体实验设计:选取6株急性髓系白血病(Acute Myeloid Leukemia, AML)细胞系,对二甲基亚砜(Dimethyl Sulfoxide, DMSO)对照组、单药处理组与联合处理组(ATRA与GSK2879552)开展基因表达分析,设置两个采样时间点,每个处理设置两次生物学重复,共计96个样本进行双端RNA测序(paired end RNA-seq)。
