Our study represents a new strategy for identifying drivers and risk factors of uterine fibroids (F) by identifying genes and pathways differentially regulated in myometrial stem cells (SCs) isolated from myometrium without fibroids (MyoN) and from myometrium adjacent to uterine fibroids (MyoF) using RNA-seq approach. Moreover, we will perform the comparison analysis of the transcriptome between MyoF SCs and fibroid SCs to identify differentially expressed genes. 本研究通过RNA测序(RNA-seq)技术,对分离自无肌瘤子宫肌层组织(MyoN)以及子宫肌瘤(F)毗邻子宫肌层组织(MyoF)的子宫肌层干细胞(myometrial stem cells, SCs)中差异调控的基因与通路展开分析,以此建立识别子宫肌瘤驱动因子与风险因素的全新研究策略。此外,本研究还将对MyoF来源干细胞与肌瘤来源干细胞的转录组(transcriptome)开展对比分析,以筛选差异表达基因(differentially expressed genes)。总体实验设计:对三类样本的人源子宫肌层干细胞的转录组进行检测,三类样本分别为:无肌瘤子宫肌层组织(MyoN)来源、子宫肌瘤毗邻子宫肌层组织(MyoF)来源,以及子宫肌瘤(F)来源。
