GRASPing experience-dependent protein expression signatures enriched for hippocampal engram cell synapses

To investigate how learning shapes the molecular composition of synapses we enriched specific synaptic subpopulations from the hippocampus using fluorescence-based tagging and flow-cytometric synapse sorting. Sorted synaptosomes were analysed using label-free quantitative mass spectrometry–based proteomics. Comparative profiling across different behavioral experiences revealed distinct protein signatures associated with synaptic structure and function.
为探究学习如何塑造突触的分子组成,我们采用荧光标记结合流式细胞术突触分选的方法,从海马体中富集了特定的突触亚群。随后,我们利用无标记定量质谱蛋白质组学技术,对分选得到的突触体进行分析。通过比对不同行为经历样本的蛋白质表达谱,我们发现了与突触结构及功能密切相关的特异性蛋白质特征。
