2nd time collection: Jan 2016. Defocus -6.Lp02 is thymidine auxotroph of the parental strain Lp01 originating from Philadelphia-1 strain. Cells were grown in AYE liquid culture with following supplements: ferric nitrate, cysteine hydrochloride and thymidine. Scrape cells off a plate, dissolve in the media, adjust the cell density to 0.6 and make a dilution series. Start O/N culture with all of them and freeze cells next day from the one in early stationary phase that is OD ~ 3.0

第二次样本采集：2016年1月。Defocus -6.Lp02为源自费城-1（Philadelphia-1）菌株的亲本菌株Lp01的胸腺嘧啶营养缺陷型菌株。菌体于添加了硝酸铁、半胱氨酸盐酸盐及胸腺嘧啶的AYE液体培养基中培养。刮取平板上的菌体，将其重悬于培养基后调整细胞密度至0.6，并制备梯度稀释系列。以各梯度稀释菌液启动过夜（O/N）培养，次日选取处于早期稳定期、光密度（Optical Density，OD）约为3.0的培养物收集菌体并冻存。
