Engineered Enucleated Mesenchymal Stem Cells Regulating Immune Microenvironment and Promoting Wound Healing

We conducted a transcriptomic analysis to evaluate DEK expression levels in the uninjured skin of healthy mice, wound tissue of healthy mice, and diabetic mice. Our findings revealed a significant upregulation of DEK in diabetic mouse wounds, with Gene Ontology (GO) analysis indicating enrichment in pathways related to neutrophil migration. Moreover, principal component analysis (PCA) results indicated that there are significant differences in gene expression between these two groups (Fig. 2D). Differential expression gene (DEG) analysis revealed 508 downregulated and 359 upregulated genes in the diabetic group compared with the health group. These downregulated genes are associated with skin, chondrocyte, and cartilage development, suggesting that the wound repair process is impaired in diabetic mice. Overall design: Comparative gene expression profiling analysis of RNA-seq data for skin tissue from uninjured skin of healthy mice, wound tissue of healthy mice, and diabetic mice.
