RNA seq on primary pancreatic cancer (KPPC;Col1pdxKO cancer-collagen1 knockout) cells growing on collagen 1 homotrimers and heterotrimers. In this study, we identify that pancreatic cancer cells produce a unique Collagen1 homotrimer variant, in contrast to the Collagen1 heterotrimer produced by normal cells (such as fibroblasts). The variant forms of Collagen1 (homotrimer and heterotrimer) have distinct effects on cancer cell behaviors. We culture the pancreatic cancer cells on purified Collagen1 homotrimer and heterotrimer (as well as vehicle control) and examine the global changes of gene expression profile in cancer cells by variant Collagen1 subtypes. Primary pancreatic cancer (KPPC;Col1pdxKO cancer-collagen1 knockout) cells were cultured in the presence of: (1) coated vehicle (2) coated Collagen 1 homotrimers (3) coated Collagen 1 heterotrimers. A total of 9 samples (3 biological replicates per group) were sequenced on Illumina Nextseq 500.

本研究针对培养于I型胶原(Collagen1)同源三聚体(homotrimer)与异源三聚体(heterotrimer)的原发性胰腺癌(KPPC;Col1pdxKO,即胶原1敲除型胰腺癌)细胞开展RNA测序(RNA-seq)分析。本研究证实,与成纤维细胞等正常细胞产生的I型胶原异源三聚体不同,胰腺癌细胞可生成独特的I型胶原同源三聚体变体。两种I型胶原变体(同源三聚体与异源三聚体)对癌细胞行为具有截然不同的调控作用。我们将胰腺癌细胞分别接种于纯化的I型胶原同源三聚体、异源三聚体(以及溶剂对照)表面,分析不同亚型I型胶原诱导的癌细胞基因表达谱全局变化。本研究使用的原发性胰腺癌(KPPC;Col1pdxKO)细胞分为三组培养条件:(1) 溶剂包被对照组;(2) I型胶原同源三聚体包被组;(3) I型胶原异源三聚体包被组。每组设置3次生物学重复,共计9个样本,均在Illumina Nextseq 500测序平台上完成测序。
