CREBBP/EP300 acetyltransferase inhibition targets lineage specific enhancers to inhibit the proliferation of ER+ breast cancer cells [RNA-Seq]

MCF7, T47D, or ZR751 cells were treated with a CREBBP/EP300 acetyltransferase inhibitor CPI-1612 and/or Fulvestrant and expression changes were measured via RNA-seq Overall design: MCF7, T47D, or ZR751 cells were treated with DMSO, CPI-1612 (5 nM, 50 nM) or Fulvestrant (100 nM), or combinations of CPI-1612 + Fulvestrant. Whole RNA was extracted and used for library construction and RNA-sequencing.

以MCF7、T47D及ZR751细胞为实验材料，使用CREBBP/EP300乙酰转移酶抑制剂CPI-1612与/或氟维司群（Fulvestrant）进行处理后，通过RNA测序（RNA-seq）检测细胞的基因表达变化情况。整体实验设计：将MCF7、T47D及ZR751细胞分别用二甲基亚砜（DMSO）、CPI-1612（5 nM、50 nM）、氟维司群（100 nM）单药，或CPI-1612与氟维司群联合处理；随后提取总RNA，构建测序文库并开展RNA测序。
