Metaphase chromosome staining was used to provide a high level overview of the pattern of histone modifications (H3K27ac, H3K27me3 and H3K4me3) at a single cell level in human lymphoblastois cells. These epigenomic banding patterns were related to various genomic features including gene and CpG island density as well as gene expression. Overall design: A 3 array study using RNA extracted from 3 separate cultures of human lymphoblastoid cells as biological replicates
本研究采用中期染色体染色(metaphase chromosome staining)技术,在单细胞水平上全面解析人淋巴母细胞中组蛋白修饰(histone modifications,包含H3K27ac、H3K27me3及H3K4me3)的分布模式。该研究得到的表观基因组条带模式与多种基因组特征存在关联,包括基因密度、CpG岛(CpG island)密度以及基因表达水平。
实验整体设计:以3份独立培养的人淋巴母细胞提取的RNA作为生物学重复(biological replicates),开展基于3个芯片阵列的研究。
