Phosphoproteomic analysis of diffuse-type gastric carcinoma cells with Met gene amplification identified Pleckstrin Homology Domain Containing A5 (PLEKHA5) as a protein that is tyrosine-phosphorylated downstream of Met. We used DNA microarray analysis to explore the signaling pathways downstream of PLEKHA5 that regulates the survival and peritoneal dissemination of Met-amplified 58As9 diffuse-type gastric carcinoma cells. Overall design: 58As9 cells were transfected with conrol or PLEKHA5 siRNA for RNA extraction and hybridization on Affymetrix microarrays.

针对携带Met基因扩增的弥漫型胃癌细胞开展磷酸化蛋白质组学分析,鉴定出含普列克底物蛋白同源域A5(Pleckstrin Homology Domain Containing A5,PLEKHA5)为Met下游发生酪氨酸磷酸化的蛋白质。我们采用DNA微阵列分析,探究调控Met扩增型58As9弥漫型胃癌细胞存活与腹膜播散的PLEKHA5下游信号通路。实验整体设计:将58As9细胞转染对照小干扰RNA(small interfering RNA,siRNA)或PLEKHA5小干扰RNA,随后提取RNA并在Affymetrix微阵列上进行杂交。
