The LOV2Kinase transgenic line was mutated with EMS to uncover suppressors of its activity in low light conditions. Following the identification of suppressor mutants, each mutant was backcrossed against the LOV2Kinase parent and subjected to deep sequencing to identify candidate SNPs that could act as suppressors.
本研究以LOV2Kinase转基因株系为实验材料,采用乙基甲磺酸(Ethyl Methanesulfonate,EMS)对其进行诱变,以筛选弱光条件下该株系活性的抑制突变体。在获得上述抑制突变体后,将每一株突变体与原始LOV2Kinase亲本株系回交,随后进行深度测序,以鉴定可作为抑制因子的候选单核苷酸多态性(Single Nucleotide Polymorphism,SNP)位点。
